Calcium and Strontium Coordination Polymers as Controlled Delivery Systems of the Anti-Osteoporosis Drug Risedronate and the Augmenting Effect of Solubilizers

Bisphosphonates (BPs) constitute a class of drugs used for the treatment calcium- and bone-related disorders, including osteoporosis, Paget’s disease, etc. The presence anionic phosphonate groups endows them with ability to act as ligands metal ions. As result, synthesis complexes or coordination polymers various structural motifs can be accomplished. In this work, 3rd generation BP drug risedronate (RIS) was combined biologically acceptable alkaline earth ions (e.g., Ca2+ Sr2+) in an effort synthesize new materials. These metal–RIS compounds operate controlled delivery systems (CDSs) when exposed appropriate experimental conditions, such low pH human stomach. CDS networks containing Sr2+ RIS were physicochemically structurally characterized evaluated their release free during acid-driven hydrolysis process. Due solubility at pH, cationic additives (linear polyethyleneimine amine-terminated polyaminoamide dendrimer) utilized solubilizers. Based on results study, there attempt correlate efficiency features these CDSs.
